Multiparticulate sustained release systems have attracted increasing attention in the last few decades since they offer distinctive advantages over single layer sustained release monolithic preparations by reducing the sudden risk of dose dumping, lowered intra- and inter- individual variability of dissolution of drug substance and increased residence time within the lower intestine. Developing multiparticulate formulation of a highly water soluble drug which provides optimum in vitro release over 24 hours is a challenging task. This task is facilitated by the inclusion of EUDRAGIT® polymers which have been widely used due to their potential applications in drug delivery systems. Among the EUDRAGIT® range of polymers, EUDRAGIT® NE 30 D is widely employed; it is the aqueous dispersion of a neutral copolymer based on ethyl acrylate and methyl methacrylate which offers pH independent sustained effect. The aim of the present work was to demonstrate the use of EUDRAGIT® NE as a sustained release polymer in formulating a multiparticulate system of a highly water soluble (572 mg/ml) drug venlafaxine1.
