ProteinChip Technology

The ProteinChip System employs a unique family of arrays carrying functional groups (e.g., hydrophilic, anion, and cation exchange, hydrophobic, IMAC, pre-activated chemistries) that mimic chromatography sorbent selectivities. For example, the Q10 array has a chemistry equivalent to the Pall BioSepra Q Ceramic HyperD sorbent.

Small crude samples (0.5 – 400 µL) from cell culture can be directly applied to a series of arrays and retained proteins analyzed based on their molecular mass using SELDI-MS (Surface Enhanced Laser Desorption Ionization — Mass Spectrometry) on the ProteinChip TOF-MS Reader in minutes. The SELDI technology quickly and accurately identifies proteins with molecules ranging in size from 300 Da to 500,000 Da.

Process Proteomics Services are implemented by dedicated experts in Pall Laboratories. Prior to starting full testing, a detailed proposal is submitted for approval. Once feasibility has been checked by a simple lab trial, the full experimental plan is implemented. After completion of the testing, a comprehensive report is provided. Expression Monitoring and Optimization Services

The ProteinChip technology enables rapid (1 hour) near real-time analysis of protein expression levels, protein integrity, and impurities directly from crude expression feedstocks, and it can be used as a tool to monitor upstream (fermentation and cell culture) conditions:

Information obtained includes:

  • Time course monitoring
  • Rapid qualification of best productivity strains
  • Optimized cell culture conditions
  • Relative quantification of expression
  • Monitoring of target protein integrity

Protein Purification Development and Optimization Service

For target protein capture, selectivity conditions are screened "on-chip" and directly transferred to traditional chromatography columns, significantly reducing chromatographic purification time and cost.

  • On-chip determination of a direct capture step for the target protein from a crude sample
  • Development of polishing steps from the eluate of the previous step
  • Transfer to column chromatography
  • Re optimization of an existing process

ProteinChip Arrays can be used to screen several different binding and wash conditions in parallel (for example, pH, ionic strength, and buffers) that mimic those applied on regular chromatography sorbents. Once purification conditions are optimized on-chip or with a unique 96-well plate microplate format, they can be transferred to traditional chromatography columns with minimal re-optimization.

  • Direct molecular mass of target protein and impurities
  • Quick selection of the best chromatography surfaces for target protein capture
  • Options for impurity removal (polishing step)
  • Direct transfer to scalable chromatography sorbent

Protein Integrity Check and Various Assays

The ProteinChip system is a rapid, sensitive and accurate analytical technology that can complement conventional analytical methods for applications such as:

  • Analysis and relative quantification of target protein, product, and process-related impurities from run to run. No need to develop specific antibody-based assays.
  • Protein integrity check: detection of proteolytic cleavages, and various post-translational modifications (phosphorylations, oxidations, glycosylation).
  • Protein identity confirmation: proteins bands from conventional gels can be excised for subsequent proteolytic digestion. Following direct tryptic digestion "on-chip" or not according to the sample, peptide fragments can be analyzed by SELDI, and protein identity confirmation is achieved by a search of protein databases.
