In the past few years, the interest in hot melt extrusion for pharmaceutical applications has increased tremendously. Common applications are e.g. the preparation of solid dispersions for solubility enhancement, sustained release pellets and transdermal patches. Although for example EUDRAGIT® E has a low glass transition temperature (50 °C), high temperatures above 100 °C have to be applied to either melt or soften the polymeric carrier or to melt the drug. As numerous drugs are heat sensitive the use of hot melt extrusion is in this case limited. Therefore, formulations were developed to apply melt extrusion also for the preparation of dosage forms containing heat sensitive drugs, proteins etc.
Melt extrusion was performed by either using a Haake Minilab (ThermoFisher, Karlsruhe, Germany) or a Micro 18 GL Pharma (Leistritz, Nürnberg, Germany). EUDRAGIT® E was supplied by Evonik Röhm GmbH, Pharma Polymers, Darmstadt, Germany, PEG 6000 by KMF Laborchemie, Lohmar, Germany, TEC by Merck, Darmstadt, Germany and stearic acid by Fluka GmbH, Buchs, Switzerland.
