Direct Compression Of Very High Dose Drug Tablets Using Extremely Compactible MCC, Ceolus™ KG-1000

There are some very high dose tablets in which the drug content is near 1000mg, such as anti-HIV drugs and multi-vitamin supplements. The size of such tablets is often so large that elderly people or small children can't easily swallow them. However, if the tablet size is reduced, hardness and friability can be insufficient due to limited space for excipients. In the case of such formulations, extremely highly compactible MCC CeolusTM KG-1000 is useful because a small amount is effective in increasing tablet hardness and improving friability. In this study, tableting properties of CeolusTM KG-1000 are studied to confirm its effectiveness in improving tablet hardness in comparison with other MCC products in direct compression using ascorbic acid, which is poorly compactible and may show insufficient tablet hardness in direct compression at very high dose.

  • KG-1000 has the lowest bulk density among MCC grades.
  • KG-1000 shows superior compactibility compared with PH-101 and other standard MCC grades.
  • KG-1000 particles have extremely large L/D value. The particles easily arrange perpendicularly to the applied force upon compaction; therefore the contact area of the MCC particles is increased. Entanglement of particles also easily occurred under compression force which provides additional compactibility.
