Long-Term Cell-Based Kinetics Using Tecan's GCM™ And The Thermo Scientific Nunc Edge Plate

Long-term cell-based assays often face limitations due to environmental instability within microplate readers. This application note introduces a solution that combines precise CO₂ regulation via the Gas Control Module (GCM) with the evaporation-minimizing design of the Thermo Scientific Nunc Edge plate. Together, they enable uninterrupted, multi-day kinetic studies directly inside the reader—eliminating the need for repeated plate transfers and avoiding data gaps. Experimental results using A431 carcinoma cells show significantly improved proliferation when CO₂ control is active, even in serum-free conditions. The system maintains stable pH and temperature, supporting consistent cell growth and reliable fluorescence-based measurements over 75 hours.
