mRNA Manufacturing With Fed-Batch In Vitro Transcription

In vitro transcription remains a central yet costly step in RNA manufacturing, particularly for applications like mRNA vaccines. Traditional batch IVT reactions often stall when certain components accumulate, capping overall yield and slowing development timelines. Explore a more efficient approach by applying the BioLector XT Microbioreactor to optimize IVT conditions at milliliter scale. With precise control of temperature, pH, mixing, and automated feeding, the system enables rapid testing of feed rates and concentrations that enhance mRNA output. Real-time pH monitoring also reveals a strong link between pH shifts and crude mRNA levels, creating a useful indicator of reaction progress. The process developed at 0.8 mL was then successfully translated to 20 mL and 200 mL runs in a GMP-ready rocking bioreactor, showing how microscale insights can confidently guide scale-up.
