RHOBUST® Downstream Processing Technology Video

DSM’s proprietary Rhobust® technology is a Second Generation Expanded Bed Adsorption (EBA) technology that provides an elegant solution for direct product capture in the first downstream process unit operation, by combining clarification and product capture into one single step. Centrifugation, depth-filtration and packed bed chromatography can be replaced by one unit operation, resulting in less preparation and process time, lower cost of goods and reduced investment costs.

In particular, the Rhobust® technology combines well with high cell density and high titer harvests, such as those achieved using DSM Biologics’ proprietary XD® technology. A comparison of the XD® and Rhobust® process to the classical process comprising fed-batch, filtration and packed bed chromatography shows improved recovery with comparable product purity and residual impurity levels. The use of the Rhobust® EBA technology for direct product capture from the XD® harvest circumvents the need for cell removal by centrifugation or depth filtration, resulting in shorter processing time and lower manufacturing costs. In addition, many other high-density, viscous feed streams, such as E. coli lysate, yeast fermentation broths or other biological sources of recombinant proteins can be efficiently processed using the Rhobust® technology for direct product capture.
