Sample Preparation Automation For Immunosuppressant Drug Analysis On The Fluent Mix And Pierce Workstation

Monitoring immunosuppressant drug levels is critical for transplant patients, but manual sample preparation for LC-MS/MS analysis can be error-prone and time-consuming. This application note presents a fully automated workflow using the Fluent® Mix and Pierce Workstation to streamline the MassTox® ONEMINUTE Test for whole blood samples. The system eliminates manual tube handling through piercing tips and barcode tracking to reduce contamination risk and improve traceability. Performance evaluations show high accuracy, reproducibility, and minimal cross-contamination across four key drugs: cyclosporin A, everolimus, sirolimus, and tacrolimus. The automated protocol supports up to 87 samples per run and integrates seamlessly with LC-MS/MS systems. Final patient sample testing confirmed consistent results with ≤5% variability.
