Scale-Up Of AAV Production In iCELLis™ Fixed-Bed Bioreactors

Scaling adherent AAV production often introduces variability that can stall development timelines and complicate tech transfer. Examine how a single rAAV5 production process performs across three fixed-bed bioreactor scales, from early development through industrial manufacturing. Using geometrically consistent systems and aligned operating strategies, the study evaluates cell growth, metabolic behavior, and viral productivity at surface areas spanning 2.65 to 333 m². Results show comparable metabolic profiles, stable cell densities at transfection, and consistent genomic titers per unit surface area across all scales. Also, gain insight into practical considerations for scaling parameters such as linear speed, media volume-to-surface-area ratios, and dissolved oxygen control.
