AAV Production In A Single-Use Bioreactor
With the tremendous growth in the use of viral vectors for vaccine and gene therapy applications, researchers must decide how to scale their manufacturing processes to deliver products that fulfill clinical and commercial needs. Here we describe how to use the Gibco™ CTS™ AAV-MAX Helper-Free AAV Production System Kit to produce adeno-associated virus (AAV) in the Thermo Scientific™ DynaDrive™ and HyPerforma™ 5:1 Single-Use Bioreactors (S.U.B.s) at the 50 L scale.
Gibco™ CTS™ Viral Production Cells 2.0 (VPC 2.0) were maintained throughout the seed train and in bioreactors in Gibco™ CTS™ Viral Production Medium containing 4 mM Gibco™ GlutaMAX™ Supplement. The cells were thawed and maintained in shake flasks as recommended in the AAV-MAX system user guide [1] before inoculating the bioreactors at low turndown to further expand the cells prior to each production run. The DynaDrive and HyPerforma 5:1 S.U.B.s were set up according to their respective user guides, and a Thermo Scientific™ HyPerforma™ G3Pro™ Bioprocess Controller was used to control each bioreactor. Samples were collected daily to assess the growth rate and health of the cells.
