Hydrophilic Interaction Liquid Chromatography (HILIC) Method Migration Part 2: Troubleshooting Peak Splitting Of Cetirizine

Regulated laboratories often employ multiple HPLC systems with varying configurations, making method transfer challenging. HILIC methods, specifically used for separating polar compounds, present unique challenges due to their sensitivity to system differences and potential for migration issues.

This study focused on the impact of autosampler design on HILIC method performance, particularly when using different wash solvent compositions. The objective was to investigate why a split peak occurred in the system suitability solution of a cetirizine hydrochloride organic impurities USP method when transferred from an Alliance iS HPLC System to a Vendor X HPLC System.

By comparing the autosampler designs and varying wash solvent compositions, this study aimed to identify how these factors contribute to chromatographic performance in HILIC methods. The findings could help laboratories optimize their methods and ensure reliable results when transferring between different HPLC systems.
