Influence Of Peptones On CHO Glycosylation Profiles
By AB Hachmann, A Gurunathan, A Fox, H Huzair, ML Pajak Lee, SD Holdread, JW Brooks

This study explores the impact of nutrient availability and complexity on the glycosylation profiles of monoclonal antibodies (mAb) produced by different CHO cell lines. The researchers found that the addition of plant-derived peptones or GlycanTune C+ led to significant changes in mAb glycosylation, enabling the modulation of glycan structures for a desired biosimilar profile. Different peptones, such as Cotton Peptone 200 UF, Wheat Peptone 100, and Soy Peptone 100, were tested, with varying results.
The study concluded that targeted supplementation of CHO cell cultures with peptones or GlycanTune supplements can improve productivity and glycosylation profiles. However, the impact of peptones can vary based on the cell line, basal medium, and feed strategy, suggesting further optimization is required.
