Scale-Up Of Anaerobic Bacteria Cultivation In A Single-Use Bioreactor (S.U.B)
The study assessed Thermo Scientific single-use bioreactors (S.U.B.s) in collaboration with Arranta Bio for culturing strict anaerobic bacterial strains. The anaerobic bacteria test method involved using S.U.B.s of various capacities, low-permeability tubing, and gas-impermeable films. Results showed successful maintenance of anaerobic conditions, with redox potential below -300 mV, indicating suitability for anaerobic cultivation. The S.U.B.s provided efficient homogenous mixing.
By utilizing single-use systems, the study demonstrated the advantages of anaerobic production, including streamlined switching between different strains of anaerobic bacteria, without the need for extensive cleaning and media sterilization processes associated with traditional stainless steel systems. This test for anaerobic bacteria confirmed the feasibility of maintaining anaerobic conditions using Thermo Scientific single-use bioreactors (S.U.B.s), ensuring efficient and contamination-free culturing of anaerobic strains.
Furthermore, the study emphasized the importance of utilizing low-permeability tubing and gas-impermeable films in anaerobic applications to prevent oxygen ingress. Thermo Scientific Aegis5-14 and CX5-14 films were shown to have minimal oxygen permeability, ensuring optimal anaerobic conditions within the S.U.B.s. The anaerobic bacteria test method confirmed the effectiveness of these measures in creating and sustaining anaerobic environments conducive to the growth and expression of bacterial strains.
