The accurate quantification of nucleic acid concentration is crucial for various downstream applications, such as transfection, cloning, PCR, and next-generation sequencing (NGS). These applications often require specific target nucleic acid concentrations to achieve optimal performance. Inaccurate quantification can lead to increased variability in downstream assays and negatively impact the quality of results. RNA is typically quantified using microplate readers by measuring absorbance at 260 nm. However, absorbance-based methods have several disadvantages, including interference from contaminants, signal contributions from nonspecific proteins and free nucleotides, and limited sensitivity.
